When confident, she's cheerful and speaks in a pleasant tone, though rather polite and not often loud. When she's feeling repressed or shrugging off insults from others, she gets quieter in tone and volume, becoming very closed off rather than openly violent. Misa's the type to stare with her killing intent rather than actively hurt, since she finds that easier to avoid trouble with.
She has minor ADD and can get distracted by either focusing or trouble focusing sometimes, depending on the situation. Assassination classes didn't give her this issue, but general lessons do, and though she can focus far more easily thanks to Koro-sensei's teaching style, she still needs to doodle sometimes to fully focus. It's a significantly dulled down habit now, but because it was a habit, she tends to draw in her free time anyway, and has a small notebook full of doodles that's easy to use on a desk or anywhere.

Assassination training shows not always in the overt, but the subtle. Since the incident with Itona Class 3-E have become dedicated and forward to the things that they want, whether it be working against someone else in school or trying their techniques, and this has heavily developed the "internal assassin". Bloodlust is very often and easily visible in their eyes, enough that when they're concentrating intensely on an "enemy", it can be felt in a way that is near tangible. During the baseball expedition match, Shindo (the star of the baseball team) outright psyched himself into a minor freak-out because as everyone was staring at him, he felt more like they were training guns on him than just watching him.
Afterwards, during their final push in the same episode where Karma and another student stood within hitting range, every single person on the field and in the stands felt it as well, to the point that it almost felt like a demonic show (with the thrown ball outright turning into a creepy reaper head in the episode). It caused a full, if brief, freakout in Shindo that lost them the game, especially when he had swung and nearly hit the two before, having just barely skimmed past them.
It's not just mental and aura that's changed, however. Dealing with Koro-sensei means that those in 3-E are incredibly sharp-eyed and able to follow things with ease; "fastballs", swings, or other motions people would consider quick look as if they're moving slowly to 3-E's eyes, allowing them to track such things in startling ways. Furthermore, their reaction times are so much higher that these "fast objects" can be dodged by just a hair's breadth, looking like a dangerous risk to others while being something fairly minor and easy to them because of the speed. This is also helped by their bodies' being honed thanks to training and attempts against Koro-sensei, meaning that most normal students -- Junior or even High -- are outclassed by them in these subjects.
Their response to "assassinations" and "killing" is now honed in, as well. When Nagisa thought about killing Takaoka in his match, the actual thing he needed to do was just touch his knife to him. However, once Karasuma told him it was "an assassination, not to show off strength", his first strike was an upward slice from below after being lightly pressed against him, meaning it would have cut into his chin at best (and his neck at worst) had he not dodged it. Though they do need to be encouraged to fully think of their situation as "I have to kill them", when this thought is fully solid, they can make potentially fatal strikes without hesitation.
Karasuma taught them all hand signals, so they can communicate as a silent squad when needed. Their training in guns has become extremely high as well and most can fire without recoil and have their own specialized gun on top of that.
He also had them constantly climbing the mountain behind the school, so they've all become skilled at scaling cliff walls, even sheer ones.
